Picking the accurate domain name

Now I have surfed the internet and found varying and contradicting opinions about this. So I need some more clarity from you guys. I guess it is better to have some of your keywords in your domain name, right? I have a doubt with the punctuation in between. Would it be better to use the domain name with the punctuations or without the punctuations? Do they make a difference or not? I would really like some clarification on this matter as all the other forums I consulted have very opposite views on this matter. What is your general opinion?
